COLNACOQ. Rapport final ArchiMer
Hess, Philipp; Geiger, Marie; Brochard, Solène; Lepretre, Thomas; Fessard, Valerie; Antignac, Jean-philippe; Dupont, Jacques; Sechet, Veronique; Vanel, Faustine; Mondeguer, Florence; Herve, Fabienne; Leborgne, Sabrina; Deslanglois, Gwenaëlle; Marshall, Lindsey; Rounds, Lucy; Guitton, Yann; Amzil, Zouher; Grovel, Olivier; Ruiz, Nicolas; Pouchus, Francois.

Jauffrais, Thierry.
This study has been conducted in order to develop the analysis of AZAs and to produce AZAs from A. spinosum culture. It also aimed at studying the effect of environmental and nutritional factors on growth and toxin production. The study also demonstrated a link between A. spinosum and the accumulation of AZAs in shellfish, followed by the clarification of the processes of accumulation, detoxification and biotransformation of AZAs into mussels. A quantitative analysis of AZAs in A. spinosum cultures was developed, and the formation and structure of the AZA methylated analogues was explained and minimised. This work also demonstrated the feasibility of a sustainable production of AZAs from A. spinosum culture and highlighted the main factors influencing...

Effect of the PSP-causing dinoflagellate, Alexandrium sp. on the initial feeding response of Crassostrea gigas ArchiMer
Wildish, David; Lassus, Patrick; Martin, Jennifer; Saulnier, Alinne; Bardouil, Michele.
The effects of toxic and non-toxic strains of Alexandrium sp. on the initial feeding responses of the Pacific oyster Crassostrea gigas were determined by continuous monitoring of the clearance rate of individual oysters. This was determined fluorometrically by loss of fluorescence of live microalgae introduced as food. Feeding oysters with toxic or non-toxic A. tamarense and toxic A. fundyense resulted in stop/start clearance behaviour. In contrast, control oysters fed a reference microalga, Isochrysis sp., known to support their growth, did not exhibit this behavior. Pacific oysters acclimated to Isochrysis sp., fed mixtures of Alexandrium/Isochrysis, showed further evidence of stop/start clearance. Exposure for periods up to 48 hours with non-toxic A....

Geiger, Marie.
The context of this study was the assessment of the safety of edible bivalve molluscs. Bivalve can indeed accumulate high concentrations of phycotoxins into their hepatopancreas, potentially leading to human intoxication outbreaks after consumption of such contaminated bivalves. In order to ensure the protection of shellfish consumers, regulated toxins are monitored using physico-chemical techniques. In parallel, emerging toxins, that can be putatively produced by microalgae or marine fungi, are monitored using the intraperitoneal mouse bioassay. However, this bioassay suffers ethical and methodological drawbacks. Thus, the objective of this work was to assess the ability of a bioassay suite to detect lipophilic toxins. This suite was composed of three...

Loyer, Sophie; Lazure, Pascal; Gentien, Patrick; Menesguen, Alain.
In order to study the occurrence of Gymnodinium mikimotoï bloom on the French Atlantic coast, a three-dimensional model of primary production was set up. The biological model was coupled to a hydrodynamic model previously developed at Ifremer, by the intermediary of transport, diffusion and heat fluxes. The cycles of three limiting elements for the phytoplankton growth were modelised: nitrogen, phosphorus and silicon. The second phase of the study aimed at reproducing the summer dinoflagellate blooms. In this new approach a model of the species Gymnodinium mikimotoï was coupled with the previous biomass model which provides the physical and chemical environments. The study focuses on the results obtained by this specific sub-model. The parametrisation was...

Cugier, Philippe.
The Bay of Seine is a coastal ecosystem submitted to a large river input (Seine river), which brings important quantities of nutrients. This nutrient enrichment leads to frequent eutrophication events and, from the beginning of the eighties, regular toxic phytoplanktonic (Dynophysis) blooms are observed. In order to study the nutrient cycles (nitrogen, phosphorus, silica) and their contribution to phytoplanktonic stock regulation, a three dimensional model has been developed, based on the coupling between an hydrodynamic part taking into account sediment dynamics (SAM-3D) and an ecological part, which accounts for the main biological compartments and processes. This model is able to reproduce the main physical and biological characteristics of the...
